for me, it's Raiders of the Lost Ark. Loved the adrenaline and the feeling I had as a younger GregMO sitting in a packed theatre the first time I experienced Indy and his whip.
The Howling. It's one of the very few movies that actually scared me when I first saw it.

A movie I just watched last night. Army of Darkness. Its one of my all time favorites. I first saw it when I was around 10, blew my mind.
While watching Psycho for the first time last fall I kept thinking how mind blowing the film must have been for movie goers back in 1960. It still holds up well but even though I'd never seen the film, it's so engrained in our culture that I felt like I had. So I wish I could watch Psycho for the first time as a 20-something in 1960.
Runner's up would be John Carpenter's The Thing and Taken (2009).
the princess bride for me. it was just a funny beautiful story and i haven't seen anything like it since.

Wow, great question!
It really got me thinking and as usual I can't name just one movie, but I can name 5!
First off, let me state that when I think "see it again for the first time" I'm imagining seeing it with completely virgirn eyes, on opening night, during it's respective year, with a packed house, none of whom have any idea what we're in for!! All these films I think during they're time would've been a proverbial filmgoing asskicking and I'm sorry I missed out!

I simply wish more people could see it with virgin eyes, with no knowledge/expectations of Michael Myers, fun under-rated flick!
